Background: Histone deacetylases (HDACs) play a central role in controlling cell cycle regulation, cell differentiation, and tissue development. HDAC1 is ubiquitously expressed and plays a role in cellular senescence, cardiomyocyte differentiation and hypertrophy, aging of the liver, myelination, and adult neurogenesis to name a few functions. The HDAC1 IP & Activity Assay Kit provides an antibody-based method to specifically immunoprecipitate the HDAC1 complex from cells & tissues and to measure HDAC1 activity fluorometrically. HDAC1 is immunoprecipitated from cell, nuclear or tissue extract(s) using HDAC1 antibody followed by capturing the complex on protein-A/G beads. The immunoprecipitated complex is incubated with the HDAC substrate. Only the deacetylated substrate is cleaved by the Developer to produce a fluorophore, which can be easily analyzed using a fluorescence plate reader.

NCBI Description

Histone acetylation and deacetylation, catalyzed by multisubunit complexes, play a key role in the regulation of eukaryotic gene expression. The protein encoded by this gene belongs to the histone deacetylase/acuc/apha family and is a component of the histone deacetylase complex. It also interacts with retinoblastoma tumor-suppressor protein and this complex is a key element in the control of cell proliferation and differentiation. Together with metastasis-associated protein-2, it deacetylates p53 and modulates its effect on cell growth and apoptosis. [provided by RefSeq, Jul 2008]
