Principle of the Assay: ASM ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-ASM antibody and an ASM-HRP conjugate. The assay sample and buffer are incubated together with ASM-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ASM concentration since ASM from samples and ASM-HRP conjugate compete for the anti-ASM antibody binding site. Since the number of sites is limited, as more sites are occupied by ASM from the sample, fewer sites are left to bind ASM-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ASM concentration in each sample is interpolated from this standard curve.
NCBI and Uniprot Product Information
NCBI Description
The protein encoded by this gene is a lysosomal acid sphingomyelinase that converts sphingomyelin to ceramide. The encoded protein also has phospholipase C activity. Defects in this gene are a cause of Niemann-Pick disease type A (NPA) and Niemann-Pick disease type B (NPB). Multiple transcript variants encoding different isoforms have been identified. [provided by RefSeq, Jul 2010]
Uniprot Description
SMPD1: Converts sphingomyelin to ceramide. Also has phospholipase C activities toward 1,2-diacylglycerolphosphocholine and 1,2-diacylglycerolphosphoglycerol. Isoform 2 and isoform 3 have lost catalytic activity. Monomer. Belongs to the acid sphingomyelinase family. 3 isoforms of the human protein are produced by alternative splicing.

Product Notes
The Human ASM smpd1 (Catalog #AAA731118) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA731118 ELISA Kit recognizes Human ASM. It is sometimes possible for the material contained within the vial of "Acid Sphingomyelinase, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
