This assay has high sensitivity and excellent specificity for detection of L-ARG1. No significant cross-reactivity or interference between L-ARG1 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between L-ARG1 and all the analogues, therefore, cross reaction may still exist in some cases.

Intended Uses: This L-ARG1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Canine L-ARG1. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: L-ARG1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-L-ARG1 antibody and an L-ARG1-HRP conjugate. The assay sample and buffer are incubated together with L-ARG1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the L-ARG1 concentration since L-ARG1 from samples and L-ARG1-HRP conjugate compete for the anti-L-ARG1 antibody binding site. Since the number of sites is limited, as more sites are occupied by L-ARG1 from the sample, fewer sites are left to bind L-ARG1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The L-ARG1 concentration in each sample is interpolated from this standard curve.

NCBI Description

Arginase catalyzes the hydrolysis of arginine to ornithine and urea. At least two isoforms of mammalian arginase exist (types I and II) which differ in their tissue distribution, subcellular localization, immunologic crossreactivity and physiologic function. The type I isoform encoded by this gene, is a cytosolic enzyme and expressed predominantly in the liver as a component of the urea cycle. Inherited deficiency of this enzyme results in argininemia, an autosomal recessive disorder characterized by hyperammonemia. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Sep 2011]
