Principle of the Assay: AQP-3 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-AQP-3 antibody and an AQP-3-HRP conjugate. The assay sample and buffer are incubated together with AQP-3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the AQP-3 concentration since AQP-3 from samples and AQP-3-HRP conjugate compete for the anti-AQP-3 antibody binding site. Since the number of sites is limited, as more sites are occupied by AQP-3 from the sample, fewer sites are left to bind AQP-3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The AQP-3 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes the water channel protein aquaporin 3. Aquaporins are a family of small integral membrane proteins related to the major intrinsic protein, also known as aquaporin 0. Aquaporin 3 is localized at the basal lateral membranes of collecting duct cells in the kidney. In addition to its water channel function, aquaporin 3 has been found to facilitate the transport of nonionic small solutes such as urea and glycerol, but to a smaller degree. It has been suggested that water channels can be functionally heterogeneous and possess water and solute permeation mechanisms. [provided by RefSeq, Aug 2011]

AQP3: Water channel required to promote glycerol permeability and water transport across cell membranes. Acts as a glycerol transporter in skin and plays an important role in regulating SC (stratum corneum) and epidermal glycerol content. Involved in skin hydration, wound healing, and tumorigenesis. Provides kidney medullary collecting duct with high permeability to water, thereby permitting water to move in the direction of an osmotic gradient. Slightly permeable to urea and may function as a water and urea exit mechanism in antidiuresis in collecting duct cells. It may play an important role in gastrointestinal tract water transport and in glycerol metabolism. Belongs to the MIP/aquaporin (TC 1.A.8) family. 2 isoforms of the human protein are produced by alternative splicing.
