Principle of the assay: ANX- IV ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ANX- IV antibody and an ANX- IV-HRP conjugate. The assay sample and buffer are incubated together with ANX- IV-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ANX- IV concentration since ANX- IV from samples and ANX- IV-HRP conjugate compete for the anti-ANX- IV antibody binding site. Since the number of sites is limited, as more sites are occupied by ANX- IV from the sample, fewer sites are left to bind ANX- IV-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ANX- IV concentration in each sample is interpolated from this standard curve.

NCBI Description

Annexin IV (ANX4) belongs to the annexin family of calcium-dependent phospholipid binding proteins. Although their functions are still not clearly defined, several members of the annexin family have been implicated in membrane-related events along exocytotic and endocytotic pathways. ANX4 has 45 to 59% identity with other members of its family and shares a similar size and exon-intron organization. Isolated from human placenta, ANX4 encodes a protein that has possible interactions with ATP, and has in vitro anticoagulant activity and also inhibits phospholipase A2 activity. ANX4 is almost exclusively expressed in epithelial cells. [provided by RefSeq, Jul 2008]

ANXA4: a calcium/phospholipid-binding protein which promotes membrane fusion and is involved in exocytosis. Annexins are a family of structurally related proteins whose common property is calcium-dependent binding to phospholipids. There are at least ten different annexins in mammalian species. Annexins do not contain signal peptides, yet some annexins (A1, A2 and A5) appear to be secreted in a physiologically regulated fashion.
