This is a TMB colorimetric sandwich ELISA kit with short assay time and quick experiment set up.
Principle
Human ANXA1 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Human ANXA1 in cell culture supernatants, serum and plasma (heparin, EDTA, citrate). It uses our proprietary Quick ELISA technology. Quick ELISA technology employs capture antibodies conjugated to an affinity tag that is recognized by the polyclonal antibody used to coat our Quick ELISA plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. The kit contains recombinant Human ANXA1 with immunogen: Expression system for standard: NS0, Immunogen sequence: A2-N346.

To measure Human ANXA1, add standards and samples to the wells, then add antibody cocktail. Wash the wells with PBS or TBS buffer, and add TMB. TMB is substrate to HRP and will be catalyzed to produce a blue color product, which changes into yellow after adding acidic stop solution. The density of the yellow product is linearly proportional to Human ANXA1 in the sample. Read the density of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Human ANXA1 in the sample.

Related Product Information for ANXA1 elisa kit
ANXA1, also known as lipocortin I or Annexin A1, is a protein that in humans is encoded by the ANXA1 gene. It is mapped to 9q21. 13. ANXA1 belongs to a family of Ca(2+)-dependent phospholipid binding proteins which have a molecular weight of approximately 35,000 to 40,000 and are preferentially located on the cytosolic face of the plasma membrane. ANXA1 protein has an apparent relative molecular mass of 40 kDa, with phospholipase A2 inhibitory activity. Lower peptide concentrations possibly found in inflammatory situations elicit Ca(2+) transients without fully activating the mitogen-activated protein kinase pathway. This causes a specific inhibition of the transendothelial migration of neutrophils and a desensitization of neutrophils toward a chemoattractant challenge. These findings identified ANXA1 peptides as novel, endogenous FPR ligands and established a mechanistic basis of ANXA1-mediated antiinflammatory effects.

NCBI Description

Annexin I belongs to a family of Ca(2+)-dependent phospholipid binding proteins which have a molecular weight of approximately 35,000 to 40,000 and are preferentially located on the cytosolic face of the plasma membrane. Annexin I protein has an apparent relative molecular mass of 40 kDa, with phospholipase A2 inhibitory activity. Since phospholipase A2 is required for the biosynthesis of the potent mediators of inflammation, prostaglandins and leukotrienes, annexin I may have potential anti-inflammatory activity. [provided by RefSeq, Jul 2008]

Uniprot Description

ANXA1: a calcium/phospholipid-binding protein with which promotes membrane fusion and is involved in endocytosis. Has anti-inflammatory properties and inhibits phospholipase A2 activity. Accumulates on internalized vesicles after EGF-stimulated endocytosis, suggesting that it may be required for a late stage in inward vesiculation. Phosphorylated by PKC, EGFR and Chak1. Phosphorylation results in loss of the inhibitory activity. Annexins are a family of structurally related proteins whose common property is calcium-dependent binding to phospholipids. There are at least ten different annexins in mammalian species. Annexins do not contain signal peptides, yet some annexins (A1, A2 and A5) appear to be secreted in a physiologically regulated fashion.

Product Notes

The Human ANXA1 anxa1 (Catalog #AAA1753953) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA1753953 ELISA Kit recognizes Human ANXA1. It is sometimes possible for the material contained within the vial of "Annexin A1, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
