Preparation and Storage

Principle of the Assay||This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti- AMACR antibody was pre-coated onto 96-well plates. And the biotin conjugated anti- AMACR antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. HRP-Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the AMACR amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of AMACR can be calculated.

AMACR: Racemization of 2-methyl-branched fatty acid CoA esters. Responsible for the conversion of pristanoyl-CoA and C27-bile acyl-CoAs to their (S)-stereoisomers. Has a role in drug metabolism as well as probably in lipid metabolism. The 2-arylpropionic acid (2-APA's) derivatives, including ibuprofen, are the most widely used anti-inflammatory analgesic cyclooxygenase inhibitors. The (-)-R-enantiomer, which is inactive in terms of cyclooxygenase inhibition, is epimerized in vivo via this enzyme to the cyclooxygenase-inhibiting (+)-S-enantiomer
