Intended Uses: This SMAalpha ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse SMAalpha. This ELISA kit for research use only!

Principle of the Assay: SMAalpha ELISA kit applies the quantitative sandwich enzyme immunoassay technique. The microtiter plate has been pre-coated with a monoclonal antibody specific for SMAalpha. Standards or samples are then added to the microtiter plate wells and SMAalpha if present, will bind to the antibody pre-coated wells. In order to quantitatively determine the amount of SMAalpha present in the sample, a standardized preparation of horseradish peroxidase (HRP)-conjugated polyclonal antibody, specific for SMAalpha are added to each well to "sandwich" the SMAalpha immobilized on the plate. The microtiter plate undergoes incubation, and then the wells are thoroughly washed to remove all unbound components. Next, substrate solutions are added to each well. The enzyme (HRP) and substrate are allowed to react over a short incubation period. Only those wells that contain SMAalpha and enzyme-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The SMAalpha concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene belongs to the actin family of proteins, which are highly conserved proteins that play a role in cell motility, structure and integrity. Alpha, beta and gamma actin isoforms have been identified, with alpha actins being a major constituent of the contractile apparatus, while beta and gamma actins are involved in the regulation of cell motility. This actin is an alpha actin that is found in skeletal muscle. Defects in this gene cause aortic aneurysm familial thoracic type 6. Multiple alternatively spliced variants, encoding the same protein, have been identified. [provided by RefSeq, Nov 2008]

Uniprot Description

ACTA2: Actins are highly conserved proteins that are involved in various types of cell motility and are ubiquitously expressed in all eukaryotic cells. Polymerization of globular actin (G-actin) leads to a structural filament (F-actin) in the form of a two-stranded helix. Each actin can bind to 4 others. Up-regulated in response to enterovirus 71 (EV71) infection. Belongs to the actin family.

Product Notes

The Rat alpha-SMA acta2 (Catalog #AAA720046) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA720046 ELISA Kit recognizes Rat alpha-SMA. It is sometimes possible for the material contained within the vial of "alpha Smooth Muscle Actin, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
