Bovine Actin Filament Associated Protein 1 ELISA Kit
Principle of the Assay: AFAP1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-AFAP1 antibody and an AFAP1-HRP conjugate. The assay sample and buffer are incubated together with AFAP1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the AFAP1 concentration since AFAP1 from samples and AFAP1-HRP conjugate compete for the anti-AFAP1 antibody binding site. Since the number of sites is limited, as more sites are occupied by AFAP1 from the sample, fewer sites are left to bind AFAP1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The AFAP1 concentration in each sample is interpolated from this standard curve.
NCBI and Uniprot Product Information
NCBI Description
The protein encoded by this gene is a Src binding partner. It may represent a potential modulator of actin filament integrity in response to cellular signals, and may function as an adaptor protein by linking Src family members and/or other signaling proteins to actin filaments. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Aug 2008]
Uniprot Description
AFAP: Can cross-link actin filaments into both network and bundle structures. May modulate changes in actin filament integrity and induce lamellipodia formation. May function as an adapter molecule that links other proteins, such as SRC and PKC to the actin cytoskeleton. Seems to play a role in the development and progression of prostate adenocarcinoma by regulating cell-matrix adhesions and migration in the cancer cells.
Protein type: Adaptor/scaffold
Chromosomal Location of Human Ortholog: 4p16
Cellular Component: focal adhesion; cytoplasm; actin cytoskeleton
Molecular Function: actin binding

Product Notes
The Bovine AFAP1 afap1 (Catalog #AAA747249) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA747249 ELISA Kit recognizes Bovine AFAP1. It is sometimes possible for the material contained within the vial of "Actin Filament Associated Protein 1, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
