Principle of the assay: ADR?1A ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ADR?1A antibody and an ADR?1A-HRP conjugate. The assay sample and buffer are incubated together with ADR?1A-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ADR?1A concentration since ADR?1A from samples and ADR?1A-HRP conjugate compete for the anti-ADR?1A antibody binding site. Since the number of sites is limited, as more sites are occupied by ADR?1A from the sample, fewer sites are left to bind ADR?1A-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ADR?1A concentration in each sample is interpolated from this standard curve.

Alpha-1-adrenergic receptors (alpha-1-ARs) are members of the G protein-coupled receptor superfamily. They activate mitogenic responses and regulate growth and proliferation of many cells. There are 3 alpha-1-AR subtypes: alpha-1A, -1B and -1D, all of which signal through the Gq/11 family of G-proteins and different subtypes show different patterns of activation. This gene encodes alpha-1D-adrenergic receptor. Similar to alpha-1B-adrenergic receptor gene, this gene comprises 2 exons and a single intron that interrupts the coding region.
