Principle of the Assay: The ELISA is based on the competitive binding enzyme immunoassaytechnique. The microtiter plate provided in this kit has been pre-coated with an antibody specific toVitamin B12, During the reaction,Vitamin B12in the sample or standard competes with a fixed amount of biotin-labeledVitamin B12 for sites on a pre-coated Monoclonal antibody specific to Vitamin B12. Excess conjugate and unbound sample or standard are washed from the plate. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Thena TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of Vitamin B12 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

This gene encodes a member of the ADAMTS (a disintegrin and metalloproteinase with thrombospondin motifs) protein family. Members of the family share several distinct protein modules, including a propeptide region, a metalloproteinase domain, a disintegrin-like domain, and a thrombospondin type 1 (TS) motif. Individual members of this family differ in the number of C-terminal TS motifs, and some have unique C-terminal domains. The encoded preproprotein is proteolytically processed to generate the mature enzyme. This enzyme contains two C-terminal TS motifs and functions as an aggrecanase that cleaves aggrecan, a major proteoglycan of cartilage, and may mediate cartilage destruction in osteoarthritis.
