Principle of the Assay: The Mouse ALK1 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse ALK1 in Cell Culture Supernates, Serum, Plasma. This assay employs an antibody specific for Mouse ALK1 coated on a 96-well plate. Standards and samples are pipetted into the wells and ALK1 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse ALK1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of ALK1 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background: Serine/threonine-protein kinase receptor R3 is an enzyme that in humans is encoded by the ACVRL1 gene. It is also known as activin receptor-like kinase 1, or ALK1. This gene encodes a type I cell-surface receptor for the TGF-beta superfamily of ligands. It shares with other type I receptors a high degree of similarity in serine-threonine kinase subdomains, a glycine- and serine-rich region (called the GS domain) preceding the kinase domain, and a short C-terminal tail. The encoded protein shares similar domain structures with other closely related ALK or activin receptor-like kinase proteins that form a subfamily of receptor serine/threonine kinases. Mutations in this gene are associated with hemorrhagic telangiectasia type 2, also known as Rendu-Osler-Weber syndrome 2.
