INTENDED USE This ACTN2 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Sheep ACTN2. This ELISA kit for research use only!

PRINCIPLE OF THE ASSAY ACTN2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ACTN2 antibody and an ACTN2-HRP conjugate. The assay sample and buffer are incubated together with ACTN2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ACTN2 concentration since ACTN2 from samples and ACTN2-HRP conjugate compete for the anti-ACTN2 antibody binding site. Since the number of sites is limited, as more sites are occupied by ACTN2 from the sample, fewer sites are left to bind ACTN2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ACTN2 concentration in each sample is interpolated from this standard curve.

NCBI Description

Alpha actinins belong to the spectrin gene superfamily which represents a diverse group of cytoskeletal proteins, including the alpha and beta spectrins and dystrophins. Alpha actinin is an actin-binding protein with multiple roles in different cell types. In nonmuscle cells, the cytoskeletal isoform is found along microfilament bundles and adherens-type junctions, where it is involved in binding actin to the membrane. In contrast, skeletal, cardiac, and smooth muscle isoforms are localized to the Z-disc and analogous dense bodies, where they help anchor the myofibrillar actin filaments. This gene encodes a muscle-specific, alpha actinin isoform that is expressed in both skeletal and cardiac muscles. Several transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, May 2013]

Defects in ACTN2 are the cause of cardiomyopathy dilated type 1AA (CMD1AA). Dilated cardiomyopathy is a disorder characterized by ventricular dilation and impaired systolic function, resulting in congestive heart failure and arrhythmia. Patients are at risk of premature death. Belongs to the alpha-actinin family.
