Applicable Applications for NAMPT active protein
Functional Assays (FN)
Reconstitution
Reconstitute with 0.25ml distilled water. Care should be taken during reconstitution as the protein may appear as a film at the bottom of the vial. MyBioSource recommends that the vial is gently mixed after reconstitution.N.B. Due to solubility reasons, it is recommended that the solution be kept at low pH..
Preparation
Buffer Solution
10mM HCl
Activity
The ED 50 was determined by the dose-dependant proliferation of the RPMI 8226 cells. The expected ED 50 for this effect is 15.0-20.0 ng/ml.
Target Species
Human
Preparation and Storage
Prior to reconstitution store at 4 degree C. After reconstitution store at -20 degree C. Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life: 3 months from date of reconstitution.
Related Product Information for NAMPT active protein
Visfatin is a putative cytokine originally identified as PBEF (Pre-B cell-enhancing factor), subsequently determined to be a nicotinamide phosphoribosyltransferase (NAMPT) enzyme and member of the NAPRTase family, involved in catalyzing the biosynthesis of NAD from nicotinamide. Visfatin is highly expressed in bone marrow, muscle and liver tissue and increases in expression in response to endotoxin, in amniotic epithelial cells following lipopolysaccharide (LPS) stimulation, and during the activation of T and B cells, dendritic cells and macrophages, suggesting a role in inflammatory processes.

NCBI Description

This gene encodes a protein that catalyzes the condensation of nicotinamide with 5-phosphoribosyl-1-pyrophosphate to yield nicotinamide mononucleotide, one step in the biosynthesis of nicotinamide adenine dinucleotide. The protein belongs to the nicotinic acid phosphoribosyltransferase (NAPRTase) family and is thought to be involved in many important biological processes, including metabolism, stress response and aging. This gene has a pseudogene on chromosome 10. [provided by RefSeq, Feb 2011]

Uniprot Description

PBEF: an ubiquitous protein originally described as a cytokine which acts on early B-lineage precursor cells. Other reports indicate that PBEF is not a cytokine-like secreted protein but an intracellular protein associated with the cell cycle. Contains a catalytically active nicotinate phosphoribosyltransferase domain, an enzyme involved in nicotinamide adenine dinucleotide (NAD) biosynthesis.

Product Notes

The NAMPT nampt (Catalog #AAA232426) is an Active Protein and is intended for research purposes only. The product is available for immediate purchase. AAA Biotech's VISFATIN can be used in a range of immunoassay formats including, but not limited to, Functional Assays (FN). Researchers should empirically determine the suitability of the NAMPT nampt for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "VISFATIN, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
