Purity/Purification
Highly Purified
~95% (SDS-PAGE). Purified by sequential chromotography.
Form/Format
Supplied as a lyophilized powder. BSA free. Reconstitute with sterile dH2O to 0.1-1mg/ml.
Biological Activity
ED50 range =0.005-0.02ng/ml, determined by the dose dependent cytolysis of mouse L-929 cells in the presence of actinomycin D. Optimal concentration for individual application should be determined by a dose response assay.
Endotoxin Level
~0.1ng/ug
Preparation and Storage
Lyophilized powder may be stored at 4 degree C for short-term only. Reconstitute to nominal volume by adding sterile dH2O and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Related Product Information for Tnf active protein
Tumor Necrosis Factor alpha (TNF-alpha) is a 17.3kD multi-functional pro-inflammatory cytokine primarily secreted by macrophages, but also produced by T and B lymphocytes, activated monocytes and fibroblasts. TNF-alpha exists in both a transmembrane and mature soluble form and has many biological properties, including activation of transcription factor NF-KappaB, the induction of apoptosis in various tumor cell lines and the stimulation of cell differentiation and proliferation under certain conditions. Studies have revealed a role for TNF-alpha in inflammation, septic shock, autoimmune disease and rheumatoid arthritis.

NCBI Description

acts as a cytokine; binds TNF receptors; plays a role in regulation of cell proliferation, induction of apoptosis, and inflammatory response [RGD, Feb 2006]

Uniprot Description

Function: Cytokine that binds to TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. It is mainly secreted by macrophages and can induce cell death of certain tumor cell lines. It is potent pyrogen causing fever by direct action or by stimulation of interleukin-1 secretion and is implicated in the induction of cachexia, Under certain conditions it can stimulate cell proliferation and induce cell differentiation. The TNF intracellular domain (ICD) form induces IL12 production in dendritic cells

By similarity.

Subunit structure: Homotrimer. Interacts with SPPL2B

By similarity.

Subcellular location: Cell membrane; Single-pass type II membrane protein

By similarity. Tumor necrosis factor, membrane form: Membrane; Single-pass type II membrane protein

By similarity. Tumor necrosis factor, soluble form: Secreted

By similarity. C-domain 1: Secreted

By similarity. C-domain 2: Secreted

By similarity.

Post-translational modification: The soluble form derives from the membrane form by proteolytic processing. The membrane-bound form is further proteolytically processed by SPPL2A or SPPL2B through regulated intramembrane proteolysis producing TNF intracellular domains (ICD1 and ICD2) released in the cytosol and TNF C-domain 1 and C-domain 2 secreted into the extracellular space

By similarity. The membrane form, but not the soluble form, is phosphorylated on serine residues. Dephosphorylation of the membrane form occurs by binding to soluble TNFRSF1A/TNFR1

By similarity.O-glycosylated; glycans contain galactose, N-acetylgalactosamine and N-acetylneuraminic acid

By similarity.

Sequence similarities: Belongs to the tumor necrosis factor family.

Product Notes

The Tnf tnf (Catalog #AAA650124) is an Active Protein and is intended for research purposes only. The product is available for immediate purchase. It is sometimes possible for the material contained within the vial of "Tumor Necrosis Factor alpha, Rat, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
