Biological Activity
Measured by the ability of the immobilized protein to support the adhesion of HEK293T human embryonic kidney cells. When 5x10er cells/well are added to human Olfactomedin-2 coated plates (1ug/ml, 100ul/well), ~50-75% will adhere after 1h at 37 degree C.

Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Reconstitute with 100ug/ml PBS. Aliquot and store at -20 degree C. Reconstituted product is stable for 6 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Olfactomedin-2 (OLFM2, OM2), also called OlfC or neuronal olfactomedin-related endoplasmic reticulum- localized-2 (Noelin-2) i s a 50kD (predicted), secreted member of the olfactomedin/noelin family of proteins. Since Noelin-2 has also been used to designate an alternately spliced form of Noelin-1, Olfactomedin-2 is the less ambiguous name. Olfactomedin domain-containing proteins are of ten found in neural tissues. Human Olfactomedin-2 expressed sequence tags have mainly been found in the brain and eye, and RNA in the cerebellum. In zebrafish, Olfactomedin-2 expression is developmentally regulated and, like Noelin-1, may play a role in neural crest cell differentiation. The human Olfactomedin-2 cDNA encodes a 20aa signal sequence and a 454 mature protein that contains two coiled-coil regions (aa58-85 and 136-193) and an Olfactomedin domain (aa194-446). Mature human Olfactomedin-2 shares 98% aa identity with rat, bovine and equine, 97% with mouse and canine, 81% with Xenopus, and 77% with zebrafish Olfactomedin-2. No alternate splice variants have been reported for Olfactomedin-2. Polymorphisms of human Olfactomedin-2, with or without additional polymorphisms in optineurin (OPTN), have been associated with a small fraction of high intraocular pressure or open-angle glaucoma cases in Japanese patients. The family member myocilin shows stronger association with these ocular disorders, and either myocilin or Noel in-1 might heterodimerize with Olfactomedin-2. Evidence suggests that Olfactomedin-2 has evolved from Noelin-1 gene duplication, and myocilin from Olfactomedin-2 gene duplication.

OLFM2: Peripherally associated with AMPAR complex. AMPAR complex consists of an inner core made of 4 pore-forming GluA/GRIA proteins (GRIA1, GRIA2, GRIA3 and GRIA4) and 4 major auxiliary subunits arranged in a twofold symmetry. One of the two pairs of distinct binding sites is occupied either by CNIH2, CNIH3 or CACNG2, CACNG3. The other harbors CACNG2, CACNG3, CACNG4, CACNG8 or GSG1L. This inner core of AMPAR complex is complemented by outer core constituents binding directly to the GluA/GRIA proteins at sites distinct from the interaction sites of the inner core constituents. Outer core constituents include at least PRRT1, PRRT2, CKAMP44/SHISA9, FRRS1L and NRN1. The proteins of the inner and outer core serve as a platform for other, more peripherally associated AMPAR constituents, including OLFM2. Alone or in combination, these auxiliary subunits control the gating and pharmacology of the AMPAR complex and profoundly impact their biogenesis and protein processing
