leptin antagonist; Mouse leptin antagonist (mutant L39A/D40A/F41A); LEP active protein
Ordering
For Research Use Only!
Purity/Purification
The purity of recombinant mouse leptin antagonist is greater than 98.0% as determined by:
(a) Gel filtration analysis.
(b) Analysis by reducing and non-reducing SDS-PAGE gel.
Form/Format
White lyophilized (freeze-dried) powder.
Sequence
The sequence of the first five N-terminal amino acids of recombinant mouse leptin antagonist was determined and was found to be Ala-Val-Pro-Ile-Gln
Sequence Length
167
Solubility
It is recommended to reconstitute the lyophilized recombinant mouse leptin antagonist in sterile water or sterile 0.4% NaHCO3 adjusted to pH 8-9, not less than 100ug/ml, which can then be further diluted with other aqueous solutions.
Protein Content
Protein quantization of recombinant mouse leptin antagonist was carried out by UV spectroscopy at 280 nm using the absorbency value of 0.201 as the extinction coefficient for a 0.1% (1mg/ml) solution at pH 8.0. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
Dimers and Aggregates
The purified recombinant mouse leptin antagonist (16K) consists of > 95% monomers as determined by gel-filtration chromatography.
Source
Escherichia Coli
Endotoxin
Less than 0.05 ng/ug (0.5 EU/ug) of mouse leptin antagonist.
Formulation
The recombinant protein was lyophilized from a concentrated (0.65mg/ml) solution with 0.003mM NaHCO3.
Biological Activity
Recombinant mouse leptin antagonist is capable of inhibiting leptin-induced proliferation of BAF/3 cells stably transfected with the long form of human leptin receptor. It also inhibits various leptin effects in several in vitro bioassays.
Preparation and Storage
Lyophilized recombinant mouse leptin antagonist although stable at room temperature for several weeks, should be stored desiccated below -20 degree C. Upon reconstitution at > 0.1 recombinant mouse leptin antagonist mg/ml and up to 2 mg/ml and filter sterilization recombinant mouse leptin antagonist can be stored at 4 degree C or even room temperature for several weeks making it suitable for long term infusion studies using osmotic pumps. At lower concentration of recombinant mouse leptin antagonist addition of a carrier protein (0.1% HSA or BSA) is suggested. Please prevent freeze-thaw cycles.

NCBI Description

This gene encodes a protein that is secreted by white adipocytes, and which plays a major role in the regulation of body weight. This protein, which acts through the leptin receptor, functions as part of a signaling pathway that can inhibit food intake and/or regulate energy expenditure to maintain constancy of the adipose mass. This protein also has several endocrine functions, and is involved in the regulation of immune and inflammatory responses, hematopoiesis, angiogenesis and wound healing. Mutations in this gene and/or its regulatory regions cause severe obesity, and morbid obesity with hypogonadism. This gene has also been linked to type 2 diabetes mellitus development. [provided by RefSeq, Jul 2008]

Uniprot Description

leptin: May function as part of a signaling pathway that acts to regulate the size of the body fat depot. An increase in the level of LEP may act directly or indirectly on the CNS to inhibit food intake and/or regulate energy expenditure as part of a homeostatic mechanism to maintain constancy of the adipose mass. Defects in LEP may be a cause of obesity (OBESITY). It is a condition characterized by an increase of body weight beyond the limitation of skeletal and physical requirements, as the result of excessive accumulation of body fat. Belongs to the leptin family.

Product Notes

The LEP lep (Catalog #AAA400035) is an Active Protein and is intended for research purposes only. The product is available for immediate purchase. The amino acid sequence is listed below: The sequence of the first five N-terminal amino acids of recombinan t mouse leptin antagonist was determined and was found to be Ala-Val-Pr o-Ile-Gln. It is sometimes possible for the material contained within the vial of "leptin antagonist, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
