Intended Uses: This ACTg1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Chicken ACTg1. This ELISA kit for research use only!

Principle of the Assay: ACTg1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ACTg1 antibody and an ACTg1-HRP conjugate. The assay sample and buffer are incubated together with ACTg1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ACTg1 concentration since ACTg1 from samples and ACTg1-HRP conjugate compete for the anti-ACTg1 antibody binding site. Since the number of sites is limited, as more sites are occupied by ACTg1 from the sample, fewer sites are left to bind ACTg1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ACTg1 concentration in each sample is interpolated from this standard curve.

NCBI Description

Actins are highly conserved proteins that are involved in various types of cell motility, and maintenance of the cytoskeleton. In vertebrates, three main groups of actin isoforms, alpha, beta and gamma have been identified. The alpha actins are found in muscle tissues and are a major constituent of the contractile apparatus. The beta and gamma actins co-exist in most cell types as components of the cytoskeleton, and as mediators of internal cell motility. Actin, gamma 1, encoded by this gene, is a cytoplasmic actin found in non-muscle cells. Mutations in this gene are associated with DFNA20/26, a subtype of autosomal dominant non-syndromic sensorineural progressive hearing loss. Alternative splicing results in multiple transcript variants.[provided by RefSeq, Jan 2011]
