Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for ACE has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any ACE present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ACE is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ACE bound in the initial step. The color development is stopped and the intensity of the color is measured.

Function: Converts angiotensin I to angiotensin II by release of the terminal His-Leu, this results in an increase of the vasoconstrictor activity of angiotensin. Also able to inactivate bradykinin, a potent vasodilator. Has also a glycosidase activity which releases GPI-anchored proteins from the membrane by cleaving the mannose linkage in the GPI moiety. Catalytic activity: Release of a C-terminal dipeptide, oligopeptide-|-Xaa-Yaa, when Xaa is not Pro, and Yaa is neither Asp nor Glu. Thus, conversion of angiotensin I to angiotensin II, with increase in vasoconstrictor activity, but no action on angiotensin II.
