Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Description: This Human Transmembrane protease serine 3 (TMPRSS3) ELISA Kit employs a two-site sandwich ELISA to quantitate TMPRSS3 in samples. An antibody specific for TMPRSS3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any TMPRSS3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TMPRSS3 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TMPRSS3 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: TMPRSS4 encodes a member of the serine protease family. Serine proteases are known to be involved in a variety of biological processes, whose malfunction often leads to human diseases and disorders. This gene was identified as a gene overexpressed in pancreatic carcinoma. The encoded protein is membrane bound with a N-terminal anchor sequence and a glycosylated extracellular region containing the serine protease domain. Multiple transcript variants encoding different isoforms have been found for this gene.A weak TMPRSS4 signal was detected in normal tissues of the gastrointestinal tract and in some of the urogenital tract but was not expressed in any other normal tissues. The authors suggested that TMPRSS4 may be important in processes involved in metastasis formation and tumor invasion.

NCBI Description

This gene encodes a protein that belongs to the serine protease family. The encoded protein contains a serine protease domain, a transmembrane domain, an LDL receptor-like domain, and a scavenger receptor cysteine-rich domain. Serine proteases are known to be involved in a variety of biological processes, whose malfunction often leads to human diseases and disorders. This gene was identified by its association with both congenital and childhood onset autosomal recessive deafness. This gene is expressed in fetal cochlea and many other tissues, and is thought to be involved in the development and maintenance of the inner ear or the contents of the perilymph and endolymph. This gene was also identified as a tumor-associated gene that is overexpressed in ovarian tumors. Alternatively spliced transcript variants have been described. [provided by RefSeq, Jan 2012]

TMPRSS3: Probable serine protease that play a role in hearing. Acts as a permissive factor for cochlear hair cells survival and activation at the onset of hearing and is required for saccular hair cell survival. Activates ENaC (in vitro). Defects in TMPRSS3 are the cause of deafness autosomal recessive type 8 (DFNB8). DFNA8 is a form of sensorineural hearing loss. Sensorineural deafness results from damage to the neural receptors of the inner ear, the nerve pathways to the brain, or the area of the brain that receives sound information. Defects in TMPRSS3 are the cause of deafness autosomal recessive type 10 (DFNB10). Belongs to the peptidase S1 family. 5 isoforms of the human protein are produced by alternative splicing.
