Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for NPM1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any NPM1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for NPM1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of NPM1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Description

This gene encodes a phosphoprotein which moves between the nucleus and the cytoplasm. The gene product is thought to be involved in several processes including regulation of the ARF/p53 pathway. A number of genes are fusion partners have been characterized, in particular the anaplastic lymphoma kinase gene on chromosome 2. Mutations in this gene are associated with acute myeloid leukemia. More than a dozen pseudogenes of this gene have been identified. Alternative splicing results in multiple transcript variants.[provided by RefSeq, Nov 2009]

It plays a role in controlling centrosome duplication, and may function in the assembly and/or transport of the ribosome. Two alternatively spliced isoforms have been described.
