Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for FSTL3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any FSTL3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for FSTL3 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of FSTL3 bound in the initial step. The color development is stopped and the intensity of the color is measured.

FSTL3: Isoform 1 or the secreted form is a binding and antagonizing protein for members of the TGF-beta family, such us activin, BMP2 and MSTN. Inhibits activin A-, activin B-, BMP2- and MSDT-induced cellular signaling; more effective on activin A than on activin B. Involved in bone formation; inhibits osteoclast differentiationc. Involved in hematopoiesis; involved in differentiation of hemopoietic progenitor cells, increases hematopoietic cell adhesion to fibronectin and seems to contribute to the adhesion of hematopoietic precursor cells to the bone marrow stroma. Isoform 2 or the nuclear form is probably involved in transcriptional regulation via interaction with MLLT10. A chromosomal aberration involving FSTL3 is found in a case of B-cell chronic lymphocytic leukemia. Translocation t(11;19)(q13;p13) with CCDN1. 2 isoforms of the human protein are produced by alternative initiation.
