GRK4 blocking peptide

GRK4 Antibody (C-term) Blocking Peptide

Gene Names
GRK4; IT11; GPRK4; GRK4a; GPRK2L
Synonyms
GRK4; GRK4 Antibody (C-term) Blocking Peptide; G protein-coupled receptor kinase 4; G protein-coupled receptor kinase GRK4; ITI1; GPRK2L; GPRK4; GRK4 blocking peptide
Ordering
Specificity
The synthetic peptide sequence used to generate the antibody was selected from the C-term region of human GRK4. A 10 to 100 fold molar excess to antibody is recommended. Precise conditions should be optimized for a particular assay.
Form/Format
The synthetic peptide was lyophilized with 100% acetonitrile and is supplied as a powder. Reconstitute with 0.1 ml deionized water for a final concentration of 1 mg/ml.
Sequence Length
578
Cellular Location
Cytoplasm. Cytoplasm, cell cortex. Note: Both at the cell surface and dispersed in the cytoplasm under basal conditions. Receptor stimulation results in the internalization of GRK4 to the perinuclear area, where colocalization with DRD3 is observed strongly at 5 and 15 minutes. DRD3 and GRK4 colocalize in lipid rafts of renal proximal tubule cells
Tissue Location
Isoform 1, isoform 2, isoform 3, and isoform 4 are expressed in testis. Isoform 4 is expressed in myometrium
Preparation and Storage
Maintain refrigerated at 2-8 degree C for up to 6 months. For long term storage store at -20 degree C.
Related Product Information for GRK4 blocking peptide
Specifically phosphorylates the activated forms of G protein-coupled receptors. GRK4-alpha can phosphorylate rhodopsin and its activity is inhibited by calmodulin; the other three isoforms do not phosphorylate rhodopsin and do not interact with calmodulin. GRK4-alpha and GRK4-gamma phosphorylate DRD3. Phosphorylates ADRB2.

NCBI Description

This gene encodes a member of the guanine nucleotide-binding protein (G protein)-coupled receptor kinase subfamily of the Ser/Thr protein kinase family. The protein phosphorylates the activated forms of G protein-coupled receptors thus initiating its deactivation. This gene has been linked to both genetic and acquired hypertension. Several transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Nov 2013]

Uniprot Description

GRK4: Specifically phosphorylates the activated forms of G protein-coupled receptors. Belongs to the protein kinase superfamily. AGC Ser/Thr protein kinase family. GPRK subfamily. 4 isoforms of the human protein are produced by alternative splicing.

Protein type: Protein kinase, Ser/Thr (non-receptor); Kinase, protein; EC 2.7.11.16; Protein kinase, AGC; AGC group; GRK family; GRK subfamily

Chromosomal Location of Human Ortholog: 4p16.3

Molecular Function: rhodopsin kinase activity

Biological Process: receptor internalization; regulation of G-protein coupled receptor protein signaling pathway; regulation of rhodopsin mediated signaling

Product Notes

The GRK4 grk4 (Catalog #AAA9226487) is a Blocking Peptide and is intended for research purposes only. The product is available for immediate purchase. It is sometimes possible for the material contained within the vial of "GRK4, Blocking Peptide" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
