Biological Significance
The 14-3-3 family of proteins plays a key regulatory role in signal transduction, checkpoint control, apoptotic and nutrient-sensing pathways. 14-3-3 proteins are highly conserved and ubiquitously expressed. There are at least seven isoforms, beta, gamma, epsilon, sigma, zeta, tau and eta that have been identified in mammals. The 14-3-3 gamma, a subtype of the 14-3-3 family of proteins, was thought to be brain and neuron-specific. It has been shown to interact with RAF1 and protein kinase C, proteins involved in various signal transduction pathways. Recombinant human YWHAG was expressed in E Coli and purified by using conventional chromatography techniques.

NCBI Description

This gene product belongs to the 14-3-3 family of proteins which mediate signal transduction by binding to phosphoserine-containing proteins. This highly conserved protein family is found in both plants and mammals, and this protein is 100% identical to the rat ortholog. It is induced by growth factors in human vascular smooth muscle cells, and is also highly expressed in skeletal and heart muscles, suggesting an important role for this protein in muscle tissue. It has been shown to interact with RAF1 and protein kinase C, proteins involved in various signal transduction pathways. [provided by RefSeq, Jul 2008]

Uniprot Description

14-3-3 gamma: Adapter protein implicated in the regulation of a large spectrum of both general and specialized signaling pathways. Binds to a large number of partners, usually by recognition of a phosphoserine or phosphothreonine motif. Binding generally results in the modulation of the activity of the binding partner. Homodimer. Interacts with SAMSN1. Interacts with RAF1, SSH1 and CRTC2/TORC2. Interacts with ABL1 (phosphorylated form); the interaction retains it in the cytoplasm. Interacts with GAB2. Interacts with MDM4 (phosphorylated); negatively regulates MDM4 activity toward TP53. Interacts with PKA-phosphorylated AANAT. Highly expressed in brain, skeletal muscle, and heart. Belongs to the 14-3-3 family.

Protein type: Adaptor/scaffold

Chromosomal Location of Human Ortholog: 7q11.23

Cellular Component: cytoplasmic vesicle membrane; focal adhesion; membrane; cytosol

Molecular Function: protein kinase C inhibitor activity; protein domain specific binding; insulin-like growth factor receptor binding; protein binding; protein kinase C binding; receptor tyrosine kinase binding

Biological Process: cellular response to insulin stimulus; regulation of neuron differentiation; apoptosis; organelle organization and biogenesis; negative regulation of protein kinase activity; regulation of synaptic plasticity; regulation of signal transduction; mitotic cell cycle; G2/M transition of mitotic cell cycle; protein targeting

Product Notes

The 14.3.3 gamma ywhag (Catalog #AAA835876) is a Recombinant Protein produced from Human and is intended for research purposes only. The product is available for immediate purchase. AAA Biotech's 14.3.3 gamma can be used in a range of immunoassay formats including, but not limited to, SDS-PAGE. Researchers should empirically determine the suitability of the 14.3.3 gamma ywhag for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. The amino acid sequence is listed below: 1-247 amino acids: MVDREQLVQK ARLAEQAERY DDMAAAMKNV TELNEPLSNE ERNLLSVAYK NVVGARRSSW RVISSIEQKT SADGNEKKIE MVRAYREKIE KELEAVCQDV LSLLDNYLIK NCSETQYESK VFYLKMKGDY YRYLAEVATG EKRATVVESS EKAYSEAHEI SKEHMQPTHP IRLGLALNYS VFYYEIQNAP EQACHLAKTA FDDAIAELDT LNEDSYKDST LIMQLLRDNL TLWTSDQQDD DGGEGNN. It is sometimes possible for the material contained within the vial of "14.3.3 gamma, Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
