Immunohistochemistry (IHC)

(Immunohistochemical analysis of PER1 staining in human stomach formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.)

Immunofluorescence (IF)/Immunocytochemistry (ICC)

(Immunofluorescent analysis of PER1 staining in U2OS cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 degree C in a hidified chamber. Cells were washed with PBST and incubated with a Alexa Fluor 488-conjugated secondary antibody (green) in PBS at room temperature in the dark.)

NCBI Description

This gene is a member of the Period family of genes and is expressed in a circadian pattern in the suprachiasmatic nucleus, the primary circadian pacemaker in the mammalian brain. Genes in this family encode components of the circadian rhythms of locomotor activity, metabolism, and behavior. This gene is upregulated by CLOCK/ARNTL heterodimers but then represses this upregulation in a feedback loop using PER/CRY heterodimers to interact with CLOCK/ARNTL. Polymorphisms in this gene may increase the risk of getting certain cancers. Alternative splicing has been observed in this gene; however, these variants have not been fully described. [provided by RefSeq, Jan 2014]

Uniprot Description

PER1: Component of the circadian clock mechanism which is essential for generating circadian rhythms. Negative element in the circadian transcriptional loop. Influences clock function by interacting with other circadian regulatory proteins and transporting them to the nucleus. Negatively regulates CLOCK|NPAS2-BMAL1|BMAL2-induced transactivation. Can bind heme. 3 isoforms of the human protein are produced by alternative splicing.

Protein type: Transcription factor

Chromosomal Location of Human Ortholog: 17p13.1

Cellular Component: cytoplasm; nucleus

Molecular Function: signal transducer activity; chromatin DNA binding; ubiquitin protein ligase binding; kinase binding; transcription factor binding

Biological Process: circadian rhythm; negative regulation of JNK cascade; response to cAMP; transcription, DNA-dependent; positive regulation of transcription from RNA polymerase II promoter; entrainment of circadian clock; regulation of sodium ion transport; circadian regulation of gene expression; negative regulation of I-kappaB kinase/NF-kappaB cascade; negative regulation of transcription from RNA polymerase II promoter; negative regulation of transcription, DNA-dependent; regulation of circadian rhythm; signal transduction; regulation of hair cycle; entrainment of circadian clock by photoperiod

Product Notes

The PER1 per1 (Catalog #AAA8307649) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-PER1 Antibody reacts with Human, Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's PER1 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunohistochemistry (IHC), Immunofluorescence (IF), Immunocytochemistry (ICC). WB: 1:500-1:2000 IHC: 1:50-1:200 IF/ICC: 1:50-1:200. Researchers should empirically determine the suitability of the PER1 per1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "PER1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
