Background/Introduction: Cluster of Differentiation 86 (also known as CD86 and B7-2) is aprotein expressed on antigen-presenting cells that provides costimulatory signals necessary for T cell activation and survival. The CD86 gene encodes a type I membrane protein that is a member of the immunoglobulin superfamily. Using fluorescence in situ hybridization mapping, the CD86, like CD80, was mapped to human 3q21 and mouse chromosome 16, band B5. The antigen presentation coactivators B71 and B72, which are important in other immune-mediated thyroid diseases, are important for lymphocytic infiltration and the immune response against thyroid carcinoma.

Principle of the Assay: The Bioscience Mouse CD86 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse CD86 in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Mouse CD86 coated on a 96-well plate. Standards and samples are pipetted into the wells and CD86 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse CD86 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of CD86 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Uniprot Description

CD86: Receptor involved in the costimulatory signal essential for T-lymphocyte proliferation and interleukin-2 production, by binding CD28 or CTLA-4. May play a critical role in the early events of T-cell activation and costimulation of naive T-cells, such as deciding between immunity and anergy that is made by T- cells within 24 hours after activation. Isoform 2 interferes with the formation of CD86 clusters, and thus acts as a negative regulator of T-cell activation. 4 isoforms of the human protein are produced by alternative splicing.
