1. Centrifuge the tube for a few seconds and ensure the pellet at the bottom of the tube.2. Re-dissolve the pellet using 200uL pure water and boil for 2-5 min.
Sequence Construction
A DNA sequence encoding the human MSRB3 (NP_001026849.1) (Gly26-Asp181) was expressed with a polyhistidine tag at the C-terminus.
Preparation Method
Cell lysate was prepared by homogenization of the over-expressed cells in ice-cold modified RIPA Lysis Buffer with cocktail of protease inhibitors. Cell debris was removed by centrifugation. Protein concentration was determined by Bradford assay (protein assay, Microplate Standard assay). The cell lysate was boiled for 5 min in 1 x SDS loading buffer (50mM Tris-HCl pH 6.8, 12.5% glycerol, 1% sodium dodecylsulfate, 0.01% bromophenol blue) containing 5% b-mercaptoethanol, and lyophilized.
Preparation and Storage
Store at 4 degree C for up to twelve months from date of receipt. After re-dissolution, aliquot and store at -80 degree C for up to twelve months. Avoid repeated freeze-thaw cycles.
Samples are stable for up to twelve months from date of receipt.

MSRB3 (Methionine Sulfoxide Reductase B3) is a Protein Coding gene. The protein encoded by this gene catalyzes the reduction of methionine sulfoxide to methionine. This enzyme acts as a monomer and requires zinc as a cofactor. This cysteine residue of MSRB3 is conserved in orthologs from yeast to humans and is involved in binding structural zinc. The affected individuals of six unrelated families were homozygous for the same transversion (c.265T>G) in MSRB3. In the inner ear, it is found in the sensory epithelium of the organ of Corti and vestibular end organs as well as in cells of the spiral ganglion. MSRB3-catalyzed reduction of methionine sulfoxides to methionine is essential for hearing. Diseases associated with MSRB3 include Deafness, Autosomal Recessive 74, and Autosomal Recessive Non-Syndromic Sensorineural Deafness Type Dfnb.

This Human MSRB3 overexpression lysate was created in HEK293 Cells and intented for use as a Western blot (WB) positive control. Purification of MSRB3 protein from the overexpression lysate was verified.

The protein encoded by this gene catalyzes the reduction of methionine sulfoxide to methionine. This enzyme acts as a monomer and requires zinc as a cofactor. Several transcript variants encoding two different isoforms have been found for this gene. One of the isoforms localizes to mitochondria while the other localizes to endoplasmic reticula. MSRB3: Catalyzes the reduction of free and protein-bound methionine sulfoxide to methionine. Isoform 2 is essential for hearing. Defects in MSRB3 are the cause of deafness autosomal recessive type 74 (DFNB74). A form of non-syndromic sensorineural deafness characterized by prelingual, bilateral, profound hearing loss. Sensorineural deafness results from damage to the neural receptors of the inner ear, the nerve pathways to the brain, or the area of the brain that receives sound information. A nonsense mutation affecting exclusively mitochondrial isoform 2 is sufficient to produce hearing loss. Belongs to the MsrB Met sulfoxide reductase family. 2 isoforms of the human protein are produced by alternative splicing.
