Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti- HBEGF antibody was pre-coated onto 96-well plates. And the biotin conjugated anti- HBEGF antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and washed with wash buffer. HRPStreptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the HBEGF amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of HBEGF can be calculated.

Uniprot Description

HBEGF: Growth factor that mediates its effects via EGFR, ERBB2 and ERBB4. Required for normal cardiac valve formation and normal heart function. Promotes smooth muscle cell proliferation. May be involved in macrophage-mediated cellular proliferation. It is mitogenic for fibroblasts, but not endothelial cells. It is able to bind EGF receptor/EGFR with higher affinity than EGF itself and is a far more potent mitogen for smooth muscle cells than EGF. Also acts as a diphtheria toxin receptor.
