Principle of the assay: GATA4 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-GATA4 antibody and an GATA4-HRP conjugate. The assay sample and buffer are incubated together with GATA4-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GATA4 concentration since GATA4 from samples and GATA4-HRP conjugate compete for the anti-GATA4 antibody binding site. Since the number of sites is limited, as more sites are occupied by GATA4 from the sample, fewer sites are left to bind GATA4-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GATA4 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of the GATA family of zinc-finger transcription factors. Members of this family recognize the GATA motif which is present in the promoters of many genes. This protein is thought to regulate genes involved in embryogenesis and in myocardial differentiation and function. Mutations in this gene have been associated with cardiac septal defects. This protein is thought to regulate genes involved in embryogenesis and in myocardial differentiation and function. May regulate a set of cardiac-specific genes and play a crucial role in cardiogenesis. Defects are a cause of atrial septal defect 2 (ASD2). ASD2 is an autosomal dominant condition with atrial septal defect and other congenital heart disease but no conduction defects or noncardiac abnormalities.
