Canine Prokineticin Receptor 2 ELISA Kit
Principle of the Assay: PKR2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-PKR2 antibody and an PKR2-HRP conjugate. The assay sample and buffer are incubated together with PKR2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PKR2 concentration since PKR2 from samples and PKR2-HRP conjugate compete for the anti-PKR2 antibody binding site. Since the number of sites is limited, as more sites are occupied by PKR2 from the sample, fewer sites are left to bind PKR2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PKR2 concentration in each sample is interpolated from this standard curve.

Prokineticins are secreted proteins that can promote angiogenesis and induce strong gastrointestinal smooth muscle contraction. The protein encoded by this gene is an integral membrane protein and G protein-coupled receptor for prokineticins. The encoded protein is similar in sequence to GPR73, another G protein-coupled receptor for prokineticins. [provided by RefSeq, Jul 2008]

PROKR2: Receptor for prokineticin 2. Exclusively coupled to the G(q) subclass of heteromeric G proteins. Activation leads to mobilization of calcium, stimulation of phosphoinositide turnover and activation of p44/p42 mitogen-activated protein kinase. Defects in PROKR2 are the cause of Kallmann syndrome type 3 (KAL3); also known as hypogonadotropic hypogonadism and anosmia. Anosmia or hyposmia is related to the absence or hypoplasia of the olfactory bulbs and tracts. Hypogonadism is due to deficiency in gonadotropin-releasing hormone and probably results from a failure of embryonic migration of gonadotropin- releasing hormone-synthesizing neurons. KAL3 patients have variable degrees of olfactory and reproductive dysfunction, but do not show any of the occasional clinical anomalies reported in Kallmann syndrome such as renal agenesis, cleft lip and/or palate, selective tooth agenesis, and bimanual synkinesis.
