Principle of the assay: ADCY4 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ADCY4 antibody and an ADCY4-HRP conjugate. The assay sample and buffer are incubated together with ADCY4-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ADCY4 concentration since ADCY4 from samples and ADCY4-HRP conjugate compete for the anti-ADCY4 antibody binding site. Since the number of sites is limited, as more sites are occupied by ADCY4 from the sample, fewer sites are left to bind ADCY4-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ADCY4 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of the family of adenylate cyclases, which are membrane-associated enzymes that catalyze the formation of the secondary messenger cyclic adenosine monophosphate (cAMP). Mouse studies show that adenylate cyclase 4, along with adenylate cyclases 2 and 3, is expressed in olfactory cilia, suggesting that several different adenylate cyclases may couple to olfactory receptors and that there may be multiple receptor-mediated mechanisms for the generation of cAMP signals. Alternative splicing results in transcript variants. [provided by RefSeq, Nov 2010]
