Principle of the Assay: CNTNAP1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CNTNAP1 antibody and an CNTNAP1-HRP conjugate. The assay sample and buffer are incubated together with CNTNAP1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CNTNAP1 concentration since CNTNAP1 from samples and CNTNAP1-HRP conjugate compete for the anti-CNTNAP1 antibody binding site. Since the number of sites is limited, as more sites are occupied by CNTNAP1 from the sample, fewer sites are left to bind CNTNAP1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CNTNAP1 concentration in each sample is interpolated from this standard curve.

NCBI Description

The gene product was initially identified as a 190-kD protein associated with the contactin-PTPRZ1 complex. The 1,384-amino acid protein, also designated p190 or CASPR for 'contactin-associated protein,' includes an extracellular domain with several putative protein-protein interaction domains, a putative transmembrane domain, and a 74-amino acid cytoplasmic domain. Northern blot analysis showed that the gene is transcribed predominantly in brain as a transcript of 6.2 kb, with weak expression in several other tissues tested. The architecture of its extracellular domain is similar to that of neurexins, and this protein may be the signaling subunit of contactin, enabling recruitment and activation of intracellular signaling pathways in neurons.
