Principle of the assay: HHM ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-HHM antibody and an HHM-HRP conjugate. The assay sample and buffer are incubated together with HHM-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the HHM concentration since HHM from samples and HHM-HRP conjugate compete for the anti-HHM antibody binding site. Since the number of sites is limited, as more sites are occupied by HHM from the sample, fewer sites are left to bind HHM-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The HHM concentration in each sample is interpolated from this standard curve.

NCBI Description

The enzyme encoded by this gene is classified as a thiolesterase and is responsible for the hydrolysis of S-lactoyl-glutathione to reduced glutathione and D-lactate. Three transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Oct 2013]

Uniprot Description

HAGH: Thiolesterase that catalyzes the hydrolysis of S-D- lactoyl-glutathione to form glutathione and D-lactic acid. Belongs to the metallo-beta-lactamase superfamily. Glyoxalase II family. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes

The Monkey HAGH hagh (Catalog #AAA745350) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA745350 ELISA Kit recognizes Monkey HAGH. It is sometimes possible for the material contained within the vial of "Hydroxyacylglutathione hydrolase, Mitochondrial, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
