Intended Uses: This IGF-1ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Goat IGF-1. This ELISA kit for research use only!

Principle of the Assay: IGF-1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-IGF-1 antibody and an IGF-1-HRP conjugate. The assay sample and buffer are incubated together with IGF-1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the IGF-1 concentration since IGF-1 from samples and IGF-1-HRP conjugate compete for the anti-IGF-1 antibody binding site. Since the number of sites is limited, as more sites are occupied by IGF-1 from the sample, fewer sites are left to bind IGF-1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The IGF-1 concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is similar to insulin in function and structure and is a member of a family of proteins involved in mediating growth and development. The encoded protein is processed from a precursor, bound by a specific receptor, and secreted. Defects in this gene are a cause of insulin-like growth factor I deficiency. Several transcript variants encoding different isoforms have been found for this gene.[provided by RefSeq, Mar 2009]

IGF1: The insulin-like growth factors, isolated from plasma, are structurally and functionally related to insulin but have a much higher growth-promoting activity. May be a physiological regulator of [1-14C]-2-deoxy-D-glucose (2DG) transport and glycogen synthesis in osteoblasts. Stimulates glucose transport in rat bone-derived osteoblastic (PyMS) cells and is effective at much lower concentrations than insulin, not only regarding glycogen and DNA synthesis but also with regard to enhancing glucose uptake. Defects in IGF1 are the cause of insulin-like growth factor I deficiency (IGF1 deficiency). IGF1 deficiency is an autosomal recessive disorder characterized by growth retardation, sensorineural deafness and mental retardation. Belongs to the insulin family. 3 isoforms of the human protein are produced by alternative splicing.
