Principle of the assay: Apo-L1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-Apo-L1 antibody and an Apo-L1-HRP conjugate. The assay sample and buffer are incubated together with Apo-L1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the Apo-L1 concentration since Apo-L1 from samples and Apo-L1-HRP conjugate compete for the anti-Apo-L1 antibody binding site. Since the number of sites is limited, as more sites are occupied by Apo-L1 from the sample, fewer sites are left to bind Apo-L1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The Apo-L1 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a secreted high density lipoprotein which binds to apolipoprotein A-I. Apolipoprotein A-I is a relatively abundant plasma protein and is the major apoprotein of HDL. It is involved in the formation of most cholesteryl esters in plasma and also promotes efflux of cholesterol from cells. This apolipoprotein L family member may play a role in lipid exchange and transport throughout the body, as well as in reverse cholesterol transport from peripheral cells to the liver. Several different transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Nov 2008]

Defects in APOL1 are the cause of focal segmental glomerulosclerosis type 4 (FSGS4). It is a renal pathology defined by the presence of segmental sclerosis in glomeruli and resulting in proteinuria, reduced glomerular filtration rate and edema. Renal insufficiency often progresses to end-stage renal disease, a highly morbid state requiring either dialysis therapy or kidney transplantation. Belongs to the apolipoprotein L family. 2 isoforms of the human protein are produced by alternative splicing.
