Reactivity
Porcine
Specificity
This assay has high sensitivity and excellent specificity for detection of TGF-beta. No significant cross-reactivity or interference between TGF-beta and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between TGF-beta and all the analogues, therefore, cross reaction may still exist in some cases.
Samples
Serum, plasma, cell culture supernatants, body fluid and tissue homogenate
Assay Type
Quantitative Competitive
Sensitivity
0.1 ng/mL
Preparation and Storage
Store all reagents at 2-8 degree C.

Intended Uses: This TGF-beta ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of PorcineTGF-beta. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: TGF-beta ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-TGF-beta antibody and an TGF-beta-HRP conjugate. The assay sample and buffer are incubated together with TGF-beta-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the TGF-beta concentration since TGF-beta from samples and TGF-beta-HRP conjugate compete for the anti-TGF-beta antibody binding site. Since the number of sites is limited, as more sites are occupied by TGF-beta from the sample, fewer sites are left to bind TGF-beta-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The TGF-beta concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes an RGD-containing protein that binds to type I, II and IV collagens. The RGD motif is found in many extracellular matrix proteins modulating cell adhesion and serves as a ligand recognition sequence for several integrins. This protein plays a role in cell-collagen interactions and may be involved in endochondrial bone formation in cartilage. The protein is induced by transforming growth factor-beta and acts to inhibit cell adhesion. Mutations in this gene are associated with multiple types of corneal dystrophy. [provided by RefSeq, Jul 2008]

Uniprot Description

betaIG-H3: Binds to type I, II, and IV collagens. This adhesion protein may play an important role in cell-collagen interactions. In cartilage, may be involved in endochondral bone formation. By TGFB1. Highly expressed in the corneal epithelium.

Product Notes

The Porcine TGFbeta tgfbi (Catalog #AAA734482) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA734482 ELISA Kit recognizes Porcine TGFbeta. It is sometimes possible for the material contained within the vial of "Transforming Growth Factor beta, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
