Intended Uses: This P-gp ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse P-gp. This ELISA kit for research use only!

Principle of the Assay: P-gp ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-P-gp antibody and an P-gp-HRP conjugate. The assay sample and buffer are incubated together with P-gp-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the P-gp concentration since P-gp from samples and P-gp-HRP conjugate compete for the anti-P-gp antibody binding site. Since the number of sites is limited, as more sites are occupied by P-gp from the sample, fewer sites are left to bind P-gp-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The P-gp concentration in each sample is interpolated from this standard curve.

NCBI Description

ABCB5 belongs to the ATP-binding cassette (ABC) transporter superfamily of integral membrane proteins. These proteins participate in ATP-dependent transmembrane transport of structurally diverse molecules ranging from small ions, sugars, and peptides to more complex organic molecules (Chen et al., 2005 [PubMed 15760339]).[supplied by OMIM, Mar 2008]

Uniprot Description

ABCB5: Plasma membrane transporter able to mediate efflux from cells of the rhodamine dye and of the therapeutic drug doxorubicin. Responsible for the resistance to doxorubicin of a subset of malignant melanomas. Belongs to the ABC transporter superfamily. ABCB family. Multidrug resistance exporter (TC 3.A.1.201) subfamily. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes

The Rat P-gp abcb5 (Catalog #AAA727712) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA727712 ELISA Kit recognizes Rat P-gp. It is sometimes possible for the material contained within the vial of "Permeability glycoprotein, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
