Human Adapter protein CIKS (TRAF3IP2) ELISA Kit
Principle of the Assay: TRAF3IP2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-TRAF3IP2 antibody and an TRAF3IP2-HRP conjugate. The assay sample and buffer are incubated together with TRAF3IP2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the TRAF3IP2 concentration since TRAF3IP2 from samples and TRAF3IP2-HRP conjugate compete for the anti-TRAF3IP2 antibody binding site. Since the number of sites is limited, as more sites are occupied by TRAF3IP2 from the sample, fewer sites are left to bind TRAF3IP2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The TRAF3IP2 concentration in each sample is interpolated from this standard curve.

This gene encodes a protein involved in regulating responses to cytokines by members of the Rel/NF-kappaB transcription factor family. These factors play a central role in innate immunity in response to pathogens, inflammatory signals and stress. This gene product interacts with TRAF proteins (tumor necrosis factor receptor-associated factors) and either I-kappaB kinase or MAP kinase to activate either NF-kappaB or Jun kinase. Several alternative transcripts encoding different isoforms have been identified. Another transcript, which does not encode a protein and is transcribed in the opposite orientation, has been identified. Overexpression of this transcript has been shown to reduce expression of at least one of the protein encoding transcripts, suggesting it has a regulatory role in the expression of this gene. TRAF3IP2: Could be involved in the activation of both NF-kappa-B via a NF-kappa-B inhibitor kinase (IKK)-dependent mechanism and stress-activated protein kinase (SAPK)/JNK. Defects in TRAF3IP2 are the cause of susceptibility to psoriasis type 13 (PSORS13). PSORS13 is a common, chronic inflammatory disease of the skin with multifactorial etiology. It is characterized by red, scaly plaques usually found on the scalp, elbows and knees. These lesions are caused by abnormal keratinocyte proliferation and infiltration of inflammatory cells into the dermis and epidermis. 4 isoforms of the human protein are produced by alternative splicing.
