Human 40S ribosomal protein S10 (RPS10) ELISA Kit
Principle of the Assay: RPS10 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-RPS10 antibody and an RPS10-HRP conjugate. The assay sample and buffer are incubated together with RPS10-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the RPS10 concentration since RPS10 from samples and RPS10-HRP conjugate compete for the anti-RPS10 antibody binding site. Since the number of sites is limited, as more sites are occupied by RPS10 from the sample, fewer sites are left to bind RPS10-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The RPS10 concentration in each sample is interpolated from this standard curve.

Together these subunits are composed of 4 RNA species and approximately 80 structurally distinct proteins. This gene encodes a ribosomal protein that is a component of the 40S subunit. The protein belongs to the S10E family of ribosomal proteins. It is located in the cytoplasm. Variable expression of this gene in colorectal cancers compared to adjacent normal tissues has been observed, although no correlation between the level of expression and the severity of the disease has been found. As is typical for genes encoding ribosomal proteins, there are multiple processed pseudogenes of this gene dispersed through the genome. Alternate splicing results in multiple transcript variants that encode the same protein. Naturally occurring read-through transcription occurs between this locus and the neighboring locus NUDT3 (nudix (nucleoside diphosphate linked moiety X)-type motif 3).[provided by RefSeq, Feb 2011]
