Intended Uses: This ADRA1B ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse ADRA1B. This ELISA kit for research use only!

Principle of the Assay||ADRA1B ELISA kit applies the competitive enzyme immunoassay technique utilizing a Polyclonal anti-ADRA1B antibody and an ADRA1B-HRP conjugate. The assay sample and buffer are incubated together with ADRA1B-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ADRA1B concentration since ADRA1B from samples and ADRA1B-HRP conjugate compete for the anti-ADRA1B antibody binding site. Since the number of sites is limited, as more sites are occupied by ADRA1B from the sample, fewer sites are left to bind ADRA1B-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ADRA1B concentration in each sample is interpolated from this standard curve.

Alpha-1-adrenergic receptors (alpha-1-ARs) are members of the G protein-coupled receptor superfamily. They activate mitogenic responses and regulate growth and proliferation of many cells. There are 3 alpha-1-AR subtypes: alpha-1A, -1B and -1D, all of which signal through the Gq/11 family of G-proteins and different subtypes show different patterns of activation. This gene encodes alpha-1B-adrenergic receptor, which induces neoplastic transformation when transfected into NIH 3T3 fibroblasts and other cell lines. Thus, this normal cellular gene is identified as a protooncogene. This gene comprises 2 exons and a single large intron of at least 20 kb that interrupts the coding region.
