This assay has high sensitivity and excellent specificity for detection of C.met. No significant cross-reactivity or interference between C.met and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between C.met and all the analogues, therefore, cross reaction may still exist in some cases.

Intended Uses: This C.met ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of RatC.met. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: C.met ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-C.met antibody and an C.met-HRP conjugate. The assay sample and buffer are incubated together with C.met-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the C.met concentration since C.met from samples and C.met-HRP conjugate compete for the anti-C.met antibody binding site. Since the number of sites is limited, as more sites are occupied by C.met from the sample, fewer sites are left to bind C.met-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The C.met concentration in each sample is interpolated from this standard curve.

Uniprot Description

Receptor tyrosine kinase that transduces signals from the extracellular matrix into the cytoplasm by binding to hepatocyte growth factor/HGF ligand. Regulates many physiological processes including proliferation, scattering, morphogenesis and survival. Ligand binding at the cell surface induces autophosphorylation of MET on its intracellular domain that provides docking sites for downstream signaling molecules. Following activation by ligand, interacts with the PI3-kinase subunit PIK3R1, PLCG1, SRC, GRB2, STAT3 or the adapter GAB1. Recruitment of these downstream effectors by MET leads to the activation of several signaling cascades including the RAS-ERK, PI3 kinase-AKT, or PLCgamma-PKC. The RAS-ERK activation is associated with the morphogenetic effects while PI3K/AKT coordinates prosurvival effects. During embryonic development, MET signaling plays a role in gastrulation, development and migration of muscles and neuronal precursors, angiogenesis and kidney formation. In adults, participates in wound healing as well as organ regeneration and tissue remodeling. Promotes also differentiation and proliferation of hematopoietic cells ().
