Intended Uses: This ADH3 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rat ADH3. This ELISA kit for research use only!

Principle of the Assay: ADH3 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-ADH3 antibody and an ADH3-HRP conjugate. The assay sample and buffer are incubated together with ADH3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ADH3 concentration since ADH3 from samples and ADH3-HRP conjugate compete for the anti-ADH3 antibody binding site. Since the number of sites is limited, as more sites are occupied by ADH3 from the sample, fewer sites are left to bind ADH3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ADH3 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of the alcohol dehydrogenase family. Members of this family metabolize a wide variety of substrates, including ethanol, retinol, other aliphatic alcohols, hydroxysteroids, and lipid peroxidation products. The encoded protein forms a homodimer. It has virtually no activity for ethanol oxidation, but exhibits high activity for oxidation of long-chain primary alcohols and for oxidation of S-hydroxymethyl-glutathione, a spontaneous adduct between formaldehyde and glutathione. This enzyme is an important component of cellular metabolism for the elimination of formaldehyde, a potent irritant and sensitizing agent that causes lacrymation, rhinitis, pharyngitis, and contact dermatitis. The human genome contains several non-transcribed pseudogenes related to this gene. [provided by RefSeq, Oct 2008]
