Intended Uses: This CaN ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rat CaN. This ELISA kit for research use only!

Principle of the Assay: CaN ELISA kit applies the quantitative sandwich enzyme immunoassay technique. The microtiter plate has been pre-coated with a monoclonal antibody specific for CaN. Standards or samples are then added to the microtiter plate wells and CaN if present, will bind to the antibody pre-coated wells. In order to quantitatively determine the amount of CaN present in the sample, a standardized preparation of horseradish peroxidase (HRP)-conjugated polyclonal antibody, specific for CaN are added to each well to "sandwich" the CaN immobilized on the plate. The microtiter plate undergoes incubation, and then the wells are thoroughly washed to remove all unbound components. Next, substrate solutions are added to each well. The enzyme (HRP) and substrate are allowed to react over a short incubation period. Only those wells that contain CaN and enzyme-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CaN concentration in each sample is interpolated from this standard curve.

Uniprot Description

CAND1: Enhances transcription from various types of promoters. Regulatory protein that interferes with the assembly of the SCF (SKP1-CUL1-F-box protein) ubiquitin ligase complex and thereby down-regulates ubiquitination of target proteins. Prevents neddylation of CUL1 by physically blocking access to the neddylation site. Disrupts interactions between CUL1 and SKP1 and between CUL1 and F-box proteins. Binds TBP. Part of a complex that contains CUL1 and RBX1. Binds unneddylated CUL1, but cannot bind CUL1 once it has been neddylated. Binds CUL2, CUL3, CUL4A, CUL4B and CUL5. Belongs to the CAND family. 3 isoforms of the human protein are produced by alternative splicing.

Protein type: Ubiquitin conjugating system; Transcription factor

Chromosomal Location of Human Ortholog: 12q14

Cellular Component: nucleoplasm; cullin-RING ubiquitin ligase complex; membrane; cytoplasm; nucleus; ubiquitin ligase complex

Molecular Function: protein binding; TATA-binding protein binding

Product Notes

The Human CAND1 cand1 (Catalog #AAA7222095) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7222095 ELISA Kit recognizes Human CAND1. It is sometimes possible for the material contained within the vial of "Cullin associated NEDD8 dissociated protein 1 (CAND1), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
