Rat diuretic hormone/vasopressin/arginine vasopressin ELISA Kit | ADH/VP/AVP elisa kit
Rat antidiuretic hormone/vasopressin/arginine vasopressin ELISA Kit
Principle of the Assay: AVP ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-AVP antibody and an AVP-HRP conjugate. The assay sample and buffer are incubated together with AVP-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the AVP concentration since AVP from samples and AVP-HRP conjugate compete for the anti-AVP antibody binding site. Since the number of sites is limited, as more sites are occupied by AVP from the sample, fewer sites are left to bind AVP-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The AVP concentration in each sample is interpolated from this standard curve.
NCBI and Uniprot Product Information
NCBI Description
This gene encodes a precursor protein consisting of arginine vasopressin and two associated proteins, neurophysin 2 and a glycopeptide, copeptin. Arginine vasopressin is a posterior pituitary hormone which is synthesized in the supraoptic nucleus and paraventricular nucleus of the hypothalamus. Along with its carrier protein, neurophysin 2, it is packaged into neurosecretory vesicles and transported axonally to the nerve endings in the neurohypophysis where it is either stored or secreted into the bloodstream. The precursor is thought to be activated while it is being transported along the axon to the posterior pituitary. Arginine vasopressin acts as a growth factor by enhancing pH regulation through acid-base transport systems. It has a direct antidiuretic action on the kidney, and also causes vasoconstriction of the peripheral vessels. This hormone can contract smooth muscle during parturition and lactation. It is also involved in cognition, tolerance, adaptation and complex sexual and maternal behaviour, as well as in the regulation of water excretion and cardiovascular functions. Mutations in this gene cause autosomal dominant neurohypophyseal diabetes insipidus (ADNDI). [provided by RefSeq, Mar 2010]
Uniprot Description
Function: Neurophysin 2 specifically binds vasopressin.Vasopressin has a direct antidiuretic action on the kidney, it also causes vasoconstriction of the peripheral vessels.
Subcellular location: Secreted.
Involvement in disease: Diabetes insipidus, neurohypophyseal (NDI) [MIM:125700]: A disease characterized by persistent thirst, polydipsia and polyuria. Affected individuals are apparently normal at birth, but characteristically develop symptoms of vasopressin deficiency during childhood.Note: The disease is caused by mutations affecting the gene represented in this entry. Product Notes
The Rat ADH/VP/AVP avp (Catalog #AAA722100) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA722100 ELISA Kit recognizes Rat ADH/VP/AVP. It is sometimes possible for the material contained within the vial of "diuretic hormone/vasopressin/arginine vasopressin, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
