Intended Uses: This CDC42 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rat CDC42. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: CDC42 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-CDC42 antibody and an CDC42-HRP conjugate. The assay sample and buffer are incubated together with CDC42-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CDC42 concentration since CDC42 from samples and CDC42-HRP conjugate compete for the anti-CDC42 antibody binding site. Since the number of sites is limited, as more sites are occupied by CDC42 from the sample, fewer sites are left to bind CDC42-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CDC42 concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is a small GTPase of the Rho-subfamily, which regulates signaling pathways that control diverse cellular functions including cell morphology, migration, endocytosis and cell cycle progression. This protein is highly similar to Saccharomyces cerevisiae Cdc 42, and is able to complement the yeast cdc42-1 mutant. The product of oncogene Dbl was reported to specifically catalyze the dissociation of GDP from this protein. This protein could regulate actin polymerization through its direct binding to Neural Wiskott-Aldrich syndrome protein (N-WASP), which subsequently activates Arp2/3 complex. Alternative splicing of this gene results in multiple transcript variants. Pseudogenes of this gene have been identified on chromosomes 3, 4, 5, 7, 8 and 20. [provided by RefSeq, Apr 2013]
