Principle of the assay: SSRP1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-SSRP1 antibody and an SSRP1-HRP conjugate. The assay sample and buffer are incubated together with SSRP1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the SSRP1 concentration since SSRP1 from samples and SSRP1-HRP conjugate compete for the anti-SSRP1 antibody binding site. Since the number of sites is limited, as more sites are occupied by SSRP1 from the sample, fewer sites are left to bind SSRP1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The SSRP1 concentration in each sample is interpolated from this standard curve.

Uniprot Description

Component of the FACT complex, a general chromatin factor that acts to reorganize nucleosomes. The FACT complex is involved in multiple processes that require DNA as a template such as mRNA elongation, DNA replication and DNA repair. During transcription elongation the FACT complex acts as a histone chaperone that both destabilizes and restores nucleosomal structure. It facilitates the passage of RNA polymerase II and transcription by promoting the dissociation of one histone H2A-H2B dimer from the nucleosome, then subsequently promotes the reestablishment of the nucleosome following the passage of RNA polymerase II. Binds specifically to single-stranded DNA and RNA with highest affinity for nucleotides G and U. The FACT complex is required for expression of Hox genes.
