Principle of the Assay: POU5F1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-POU5F1 antibody and an POU5F1-HRP conjugate. The assay sample and buffer are incubated together with POU5F1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the POU5F1 concentration since POU5F1 from samples and POU5F1-HRP conjugate compete for the anti-POU5F1 antibody binding site. Since the number of sites is limited, as more sites are occupied by POU5F1 from the sample, fewer sites are left to bind POU5F1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The POU5F1 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a transcription factor containing a POU homeodomain that plays a key role in embryonic development and stem cell pluripotency. Aberrant expression of this gene in adult tissues is associated with tumorigenesis. This gene can participate in a translocation with the Ewing's sarcoma gene on chromosome 21, which also leads to tumor formation. Alternative splicing, as well as usage of alternative AUG and non-AUG translation initiation codons, results in multiple isoforms. One of the AUG start codons is polymorphic in human populations. Related pseudogenes have been identified on chromosomes 1, 3, 8, 10, and 12. [provided by RefSeq, Oct 2013]

Forms a trimeric complex with SOX2 on DNA and controls the expression of a number of genes involved in embryonic development such as YES1, FGF4, UTF1 and ZFP206. Critical for early embryogenesis and for embryonic stem cell pluripotency. Interacts with UBE2I and ZSCAN10. Interacts with PKM2. Interacts with WWP2. Transcriptional activity is positively regulated by PKM2. Expressed in developing brain. Highest levels found in specific cell layers of the cortex, the olfactory bulb, the hippocampus and the cerebellum. Low levels of expression in adult tissues.
