Human Choline/ethanolamine kinase (CHKB) ELISA Kit
Principle of the Assay: CHKB ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-CHKB antibody and an CHKB-HRP conjugate. The assay sample and buffer are incubated together with CHKB-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CHKB concentration since CHKB from samples and CHKB-HRP conjugate compete for the anti-CHKB antibody binding site. Since the number of sites is limited, as more sites are occupied by CHKB from the sample, fewer sites are left to bind CHKB-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CHKB concentration in each sample is interpolated from this standard curve.

Choline kinase (CK) and ethanolamine kinase (EK) catalyze the phosphorylation of choline/ethanolamine to phosphocholine/phosphoethanolamine. This is the first enzyme in the biosynthesis of phosphatidylcholine/phosphatidylethanolamine in all animal cells. The highly purified CKs from mammalian sources and their recombinant gene products have been shown to have EK activity also, indicating that both activities reside on the same protein. The choline kinase-like protein encoded by CHKL belongs to the choline/ethanolamine kinase family; however, its exact function is not known. Read-through transcripts are expressed from this locus that include exons from the downstream CPT1B locus. [provided by RefSeq, Jun 2009]
