Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for MMP-1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MMP-1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MMP-1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MMP-1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Uniprot Description

Function: Cleaves collagens of types I, II, and III at one site in the helical domain. Also cleaves collagens of types VII and X.

Catalytic activity: Cleavage of the triple helix of collagen at about three-quarters of the length of the molecule from the N-terminus, at 775-Gly-|-Ile-776 in the alpha-1(I) chain. Cleaves synthetic substrates and alpha-macroglobulins at bonds where P1' is a hydrophobic residue.

Cofactor: Binds 4 calcium ions per subunit

Enzyme regulation: Can be activated without removal of the activation peptide.
