Application Notes
Research Application(s): ELISA Detection: JES5-16E3 monoclonal antibody is useful as a detection antibody in a sandwich ELISA for quantifying mouse IL-10 protein levels. Biotinylated JES5-16E3 antibody should be paired with purified JES5-2A5 antibody as the capture antibody, with purified recombinant mouse IL-10 as the standard.
Immunofluorescence/Flow Cytometry: JES6-16E3 antibodies are useful for intracytoplasmic staining and flow cytometric analysis to identify and enumerate IL-10-positive cells within mixed cell populations. Recombinant mouse IL-10, unlabeled or biotinylated Rat IgG2b should be used as a specificity control. FITC conjugated JES5-16E3 antibodies are useful for intracellular staining in cells producing IL-10. our intracellular staining buffers are recommended for this application.
Neutralization: The LE/AF form of JES5-16E3 can be used for neutralizing mouse IL-10 bioactivity. A suitable LE/AF Rat IgG2b should be used as an isotype control.
Since applications vary, each investigator should determine the optimal concentration appropriate for individual applications.

E. coli-expressed recombinant mouse IL-10
Characterization
To ensure acceptable performance, each batch of product is tested in a sandwich ELISA to conform to characteristics of a standard reference reagent.
Preparation and Storage
The purified (UNLB) antibody is supplied as 0.5 mg purified immunoglobulin in 1.0 mL of 100 mM borate buffered saline, pH 8.2. No preservatives or amine-containing buffer salts added. Store at 2- 8 degree C.
The fluorescein (FITC) conjugate is supplied as 100 tests in 1.0 mL of PBS/NaN3 and a stabilizing agent. Store at 2-8 degree C.
The biotin (BIOT) conjugate is supplied as 0.5 mg labeled antibody in 1.0 mL of PBS/NaN3. Store at 2-8 degree C.
The low endotoxin, azide-free (LE/AF) antibody is supplied as 0.5 mg purified immunoglobulin in 1.0 mL of PBS. Aliquot and store at or below -20 degree C.
Protect conjugated forms from light. Aliquot and freeze the low endotoxin, azide-free product at -20 degree C immediately upon receipt. Each reagent is stable for the period shown on the bottle label if stored as directed.

Researchers should empirically determine the suitability of the IL-10 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "IL-10, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
